Yes you can backup all database files in one backup procedure.

Click BackUp All Databases in the Backup Options window.

Warning: When you restore all your databases from one backup file all your databases are going to be overwritten with the content of the backup file.

Note: this procedure is useful when you want to move/transfer several databases to a new computer.
